The mechanism of action of methanolic extract of Rumex alveolatus on the membranes of Candida albicans, Candida paraposilosis and Candida glabratata

Background: Candidiasis is a common fungal disease that is caused by different species of genital herpes and involves various parts of the body. These infections can be fatal in light of the host's immune status. Following the advancement of science and the importance of the health sector in societies, the use of chemical drugs has increased. However, due to side effects, high prices and the complex process of production of chemical drugs, medicinal plants have been considered as possible treatments. The aim of this study was to investigate the mechanism of action of methanolic extract of Rumex alveolatus on membranes of some Candida species. Materials and Methods: First, the methanolic extracts of the stem and leaves of Roumex alveolatus were prepared by the Soxhlet method. The antifungal effect of methanolic extract was determined by the Wells and MIC method. In order to compare the mechanism of the effect of this plant with amphotericin B, the glucose value was determined by an autoanalyzer, sodium and potassium levels using a flame photometer apparatus, and amino acids were identified by HPLC in a medium containing extract of plant and candida. Results: The highest diameter of zone of inhibition of Candida yeast was methanol extract at a concentration of 250 mg / ml. The results showed that the mean concentration of sodium, potassium and glucose in different species of Candida was significantly different from that of methanol extract compared to amphotericin B. Most of the free amino acids were glutamine, threonine, and alanine. Conclusion: This herbal extract has a mechanism similar to that of amphotericin B and could be a suitable alternative for high-dose chemical drugs. However, the potential side effects of the plant need to be checked.

ﺑﯿﻤﺎري ﻗﺎرﭼﯽ ﺷﺎﯾﻊ اﺳﺖ ﮐﻪ ﺑﻪ وﺳﯿﻠﻪ ي ﮔﻮﻧﻪ ﻫﺎي ﻣﺨﺘﻠﻒ ﺟﻨﺲ ﮐﺎﻧﺪﯾﺪا اﯾﺠﺎد ﻣﯽ ﺷﻮد و ﻗﺴﻤﺖ ﻫﺎي ﻣﺨﺘﻠـﻒ ﺑـﺪن‬ ‫را درﮔﯿﺮ ﻣﯽ ﻧﻤﺎﯾﺪ‪ .‬اﯾﻦ ﻋﻔﻮﻧﺖ ﻫﺎ ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ وﺿﻌﯿﺖ ﺳﯿﺴﺘﻢ اﯾﻤﻨﯽ ﻣﯿﺰﺑﺎن ﻣﯽ ﺗﻮاﻧﺪ در ﺣﺪ ﻣﻬﻠﮏ ﭘـﯿﺶ روﻧـﺪ‪ .‬ﺑـﻪ دﻧﺒـﺎل ﭘﯿﺸـﺮﻓﺖ ﻋﻠـﻢ و‬ ‫اﻫﻤﯿﺖ ﺑﺨﺶ ﺳﻼﻣﺖ در ﺟﻮاﻣﻊ‪ ،‬اﺳﺘﻔﺎده از داروﻫﺎي ﺷﯿﻤﯿﺎﯾﯽ اﻓﺰاﯾﺶ ﯾﺎﻓﺘﻪ اﺳﺖ و ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ ﻋﻮارض ﺟﺎﻧﺒﯽ‪ ،‬ﻗﯿﻤﺖ ﺑﺎﻻ و ﻣﺮاﺣـﻞ ﭘﯿﭽﯿـﺪه ي‬ ‫ﺗﻮﻟﯿﺪ داروﻫﺎي ﺷﯿﻤﯿﺎﯾﯽ‪ ،‬ﮔﯿﺎﻫﺎن داروﯾﯽ ﻣﻮرد ﺗﻮﺟﻪ ﻗﺮار ﮔﺮﻓﺘﻪ اﻧﺪ‪ .‬ﻫﺪف اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﺑﺮرﺳﯽ ﻣﮑﺎﻧﯿﺴـﻢ اﺛـﺮ ﻋﺼـﺎره ﻣﺘـﺎﻧﻮﻟﯽ ﮔﯿـﺎه ر وﻣﮑ ـﺲ‬ ‫آﻟﻮﺋﻮﻻﺗﻮس ﺑﺮ ﻏﺸﺎء ﺑﺮﺧﯽ ﮔﻮﻧﻪ ﻫﺎي ﮐﺎﻧﺪﯾﺪا اﺳﺖ‪.‬‬ ‫ﻣﻮاد و روشﻫﺎ‪ :‬اﺑﺘﺪا ﻋﺼﺎره ﻣﺘﺎﻧﻮﻟﯽ ﺳﺎﻗﻪ و ﺑﺮگ ﮔﯿﺎه روﻣﮑﺲ آﻟﻮﺋﻮﻻﺗﻮس ﺑﺎ روش ﺳﻮﮐﺴﻠﻪ ﺗﻬﯿﻪ ﺷﺪ‪ .‬ﺳﭙﺲ اﺛﺮ ﺿﺪ ﻗﺎرﭼﯽ ﻋﺼﺎره ﻣﺘـﺎﻧﻮﻟﯽ‬ ‫ﺑﺎ روش اﻧﺘﺸﺎر در ﭼﺎﻫﮏ و ‪ MIC‬ﺗﻌﯿﯿﻦ ﺷﺪ‪ .‬ﺑﻪ ﻣﻨﻈﻮر ﻣﻘﺎﯾﺴﻪ ﻣﮑﺎﻧﯿﺴﻢ اﺛﺮ اﯾﻦ ﮔﯿﺎه ﺑﺎ آﻣﻔﻮﺗﺮﯾﺴﯿﻦ ‪ ،B‬ﻣﻘﺪار ﮔﻠﻮﮐﺰ ﺑـﺎ دﺳـﺘﮕﺎه اﺗﻮآﻧـﺎﻟﯿﺰور‪،‬‬ ‫ﺳﺪﯾﻢ و ﭘﺘﺎﺳﯿﻢ ﺑﺎ دﺳﺘﮕﺎه ﻓﻠﯿﻢ ﻓﺘﻮﻣﺘﺮي و اﺳﯿﺪآﻣﯿﻨﻪ ﺑﺎ روش ‪ HPLC‬در ﻣﺤﯿﻂ ﺣﺎوي ﻋﺼﺎره ﮔﯿﺎه و ﻗﺎرچ ﺗﻌﯿﯿﻦ ﺷﺪ‪.‬‬ ‫ﯾﺎﻓﺘﻪﻫﺎ‪ :‬ﺑﯿﺸﺘﺮﯾﻦ ﻗﻄﺮ ﻫﺎﻟﻪ ﻋﺪم رﺷﺪ ﻣﺨﻤﺮ ﮐﺎﻧﺪﯾﺪا ﺗﻮﺳﻂ ﻋﺼﺎره ﻣﺘﺎﻧﻮﻟﯽ در ﻏﻠﻈﺖ ‪ 250‬ﻣﯿﻠﯽ ﮔﺮم ﺑﺮ ﻣﯿﻠﯽ ﻟﯿﺘﺮ ﺑـﻮد‪ .‬ﻧﺘـﺎﯾﺞ ﻧﺸـﺎن داد ﮐـﻪ‬ ‫ﻣﯿﺎﻧﮕﯿﻦ ﻏﻠﻈﺖ ﺳﺪﯾﻢ‪ ،‬ﭘﺘﺎﺳﯿﻢ و ﮔﻠﻮﮐﺰ در ﮔﻮﻧﻪ ﻫﺎي ﻣﺨﺘﻠﻒ ﮐﺎﻧﺪﯾﺪا ﺑﺮ اﺛﺮ ﻋﺼﺎره ﻣﺘﺎﻧﻮﻟﯽ در ﻣﻘﺎﯾﺴﻪ ﺑﺎ آﻣﻔﻮﺗﺮﯾﺴﯿﻦ ‪ B‬ﺗﻐﯿﯿـﺮ ﻣﻌﻨـﺎداري دارد‪.‬‬ ‫ﺑﯿﺸﺘﺮﯾﻦ اﺳﯿﺪآﻣﯿﻨﻪ ﻫﺎي آزاد ﺷﺪه ﮔﻠﻮﺗﺎﻣﯿﻦ‪ ،‬ﺗﺮه اوﻧﯿﻦ و آﻻﻧﯿﻦ ﺑﻮدﻧﺪ‪.‬‬ ‫ﺑﺤﺚ و ﻧﺘﯿﺠﻪﮔﯿﺮي‪ :‬اﯾﻦ ﻋﺼﺎره ﮔﯿﺎﻫﯽ ﻣﮑﺎﻧﯿﺴﻢ ﻣﺸﺎﺑﻬﯽ ﺑﺎ ﻣﮑﺎﻧﯿﺴﻢ آﻣﻔﻮﺗﺮﯾﺴﯿﻦ ‪ B‬دارد و ﻣﯽ ﺗﻮاﻧﺪ ﺟﺎﯾﮕﺰﯾﻦ ﻣﻨﺎﺳـﺒﯽ ﭼﻬﺎرﻣﯿﻦ ﻋﺎﻣﻞ ﻋﻔﻮﻧﺖ ﻫﺎي ﺧﻮﻧﯽ ﺑﯿﻤﺎران ﺑﺴـﺘﺮي‬ ‫اﺛــﺮات درﻣــﺎﻧﯽ آن ﻫــﺎ ﻗﺎﺑــﻞ ﺗﻮﺟــﻪ اﺳــﺖ‪.‬ﮔﯿﺎﻫﺎن ﺗﻮاﻧــﺎﯾﯽ‬ ‫در ﺑﯿﻤﺎرﺳﺘﺎن ﻫﺎ ﻣﻄﺮح ﺷﺪه اﻧﺪ ﮐﻪ ﻣﺴـﺌﻮل ﺗﻘﺮﯾﺒـﺎ ‪% 40‬‬ ‫ﻧﺎﻣﺤﺪود ﺑﺮاي ﺳﻨﺘﺰ ﺗﺮﮐﯿﺒﺎت آروﻣﺎﺗﯿﮏ دارﻧﺪ‪ ،‬ﺑﯿﺸـﺘﺮ اﯾـﻦ‬ ‫ﻣﻮارد ﻣﺮگ وﻣﯿﺮ در ﺑﯿﻤﺎرﺳﺘﺎن ﻫﺎي آﻣﺮﯾﮑﺎ ﻣﯽ ﺑﺎﺷﻨﺪ )‪.(2‬‬ ‫ﻣﻮاد از ﺗﺮﮐﯿﺒﺎت ﻓﻨﻠﯽ و ﯾﺎ ﻣﺸـﺘﻘﺎت آن ﻫـﺎ ﻫﺴـﺘﻨﺪ‪ .‬اﯾـﻦ‬ ‫اﯾﻦ ﻗﺎرچ ﻫﺎ در اﻓﺮاد ﺑﺎ ﺳﯿﺴﺘﻢ اﯾﻤﻨﯽِ ﺗﻀـﻌﯿﻒ ﺷـﺪه ‫ﮔﺮﻓﺘﻪ و ﺑﻄﻮر وﺳـﯿﻊ ﺑـﺮاي درﻣـﺎن ﻋﻔﻮﻧـﺖ ﻫـﺎي ﻗـﺎرﭼﯽ‬ ‫ﻣﻮﺿﻌﯽ‪ ،‬ﺑﺮﺧﯽ از ﺑﯿﻤﺎري ﻫـﺎي ﻏـﺪد و ﻋﻮاﻣـﻞ دﺧﯿـﻞ در‬ ‫ﺳﯿﺴﺘﻤﯿﮏ ﺑﻌـﺪ از ﭘﯿﻮﻧـﺪ‪ ،‬اﯾـﺪز‪ ،‬ﺑﯿﻤـﺎري ﻫـﺎي وﯾﺮوﺳـﯽ‪،‬‬ ‫اﯾﺠﺎد ﺿﻌﻒ اﯾﻤﻨﯽ ﺳﻠﻮﻟﯽ ﻣﻤﮑﻦ اﺳﺖ ﺑﺎﻋﺚ ﺷﻮﻧﺪ ﮐﻪ اﯾـﻦ‬ ‫اﻧﮕﻠﯽ‪ ،‬ﭘﺮﯾﻮﻧﯽ و ﻧﯿﺰ ﺷﯿﻤﯽ درﻣﺎﻧﯽ ﺑﮑﺎر ﺑﺮده ﻣـﯽ ﺷـﻮد در‬ ‫ﻣﺨﻤﺮ ﻫﺎ‪ ،‬ﺑﯿﻤﺎري زا ﮔﺮدد )‪ .(2‬ﺑﻪ دﻧﺒـﺎل ﭘﯿﺸـﺮﻓﺖ ﻋﻠـﻢ و‬ ‫اﻏﻠﺐ ﻣﻮارد ﺑﺮاي دارو درﻣـﺎﻧﯽ اوﻟﯿـﻪ‪ ،‬ﻗﺒـﻞ از ﺗﺠـﻮﯾﺰ ﯾـﮏ‬ ‫اﻫﻤﯿﺖ ﺑﺨﺶ ﺳﻼﻣﺖ در ﺟﻮاﻣﻊ‪ ،‬اﺳﺘﻔﺎده از داروﻫﺎ اﻓﺰاﯾﺶ‬ ‫آزول ﺑﻪ ﮐﺎر ﻣـﯽ رود )‪ .(5‬آﻣﻔﻮﺗﺮﯾﺴـﯿﻦ ‪ B‬ﺑـﺎوﺟﻮد اﯾﻨﮑـﻪ‬ ‫ﯾﺎﻓﺘﻪ اﺳﺖ و ﺑـﺎ ﺗﻮﺟـﻪ ﺑـﻪ ﻋـﻮارض ﺟـﺎﻧﺒﯽ‪ ،‬ﻗﯿﻤـﺖ ﺑـﺎﻻ و‬ ‫درﻣﺎﻧﯽ اﺳﺘﺎﻧﺪارد ﺑﺮاي ﻋﻔﻮﻧﺖ ﻫﺎي ﺟﺪي ﻣﺮﺑﻮط ﺑـﻪ ﮔﻮﻧـﻪ‬ ‫ﻣﺮاﺣــﻞ ﭘﯿﭽﯿــﺪه ي ﺗﻮﻟﯿــﺪ داروﻫــﺎي ﺷــﯿﻤﯿﺎﯾﯽ‪ ،‬ﮔﯿﺎﻫــﺎن‬ ‫ﻫﺎي ﮐﺎﻧﺪﯾﺪا ﻣﺤﺴﻮب ﻣﯽ ﺷﻮد‪ ،‬وﻟﯽ ﻣﻤﮑﻦ اﺳﺖ ﺑﺎ ﺳﻤّﯿﺖ‬ ‫داروﯾﯽ ﻣـﻮرد ﺗﻮﺟـﻪ ﻗـﺮار ﮔﺮﻓﺘـﻪ اﻧـﺪ‪ .‬ﻗـﺪﻣﺖ اﺳـﺘﻔﺎده از‬ ‫ﻗﺎﺑﻞ ﺗﻮﺟﻬﯽ ﻫﻤﺎﻧﻨﺪ ﺳﻤّﯿﺖ ﮐﻠﯿﻮي ﻫﻤﺮاه ﺑﺎﺷـﺪ )‪ .(6‬دﻓـﻊ‬ ‫ﮔﯿﺎﻫﺎن داروﯾﯽ ﺑﻪ ﺳﺎل ﻫﺎ ﭘﯿﺶ و ﻣﺘﻨﺎﺳﺐ ﺑﺎ ﻗﺪﻣﺖ ﺗﻤﺪن‬ ‫اﯾﻦ دارو اﻏﻠﺐ از ﻃﺮﯾﻖ ﻣﺘﺎﺑﻮﻟﯿﺴـﻢ آﻫﺴـﺘﻪ ﮐﺒـﺪي اﺳـﺖ؛‬ ‫ﺑﺮ ﻣﯽ ﮔﺮدد‪ .‬ﮔﯿﺎه درﻣﺎﻧﯽ ﺷﺎﺧﻪ اي از ﻃﺐ ﺳﻨﺘﯽ اﺳﺖ ﮐﻪ‬ ‫ﻧﯿﻤﻪ ﻋﻤﺮ در ﺣﺪود ‪ 2‬ﻫﻔﺘﻪ ﻣﯽ ﺑﺎﺷﺪ‪ .‬ﻣﻘﺪار اﻧـﺪﮐﯽ از دارو‬ ‫در ﮐﺸﻮرﻫﺎي ﺑﺎ ﺳﺎﺑﻘﻪ اي ﻣﺜﻞ اﯾﺮان ﮐﻪ ﻏﻨﯽ از ﮔﻨﺠﯿﻨﻪ ي‬ ‫از راه ادرار دﻓﻊ ﻣـﯽ ﺷـﻮد و ﺗﻌـﺪﯾﻞ دوز ﻓﻘـﻂ در اﺧـﺘﻼل‬ ‫ﮔﯿﺎﻫﺎن ﻣﺨﺘﻠﻒ ﺑﻮﻣﯽ اﺳﺖ ﺗـﺎ ﯾـﮏ ﻗـﺮن ﭘـﯿﺶ و ﻗﺒـﻞ از‬ ‫ﮐﻠﯿﻮي ﺷﺪﯾﺪ ﻻزم اﺳﺖ )‪ .(7‬ﮔﻮﻧﻪ ﻫﺎي روﻣﮑﺲ ﻣﺘﻌﻠﻖ ﺑـﻪ‬ ‫ﭘﯿــﺪاﯾﺶ داروﻫــﺎي ﺳــﻨﺘﺘﯿﮏ ﻧﻘــﺶ اﺻــﻠﯽ را در درﻣــﺎن‬ ‫ﺧﺎﻧﻮاده ي ﭘﻠﯽ ﮔﻮﻧﺎﺳﻪ )ﻋﻠﻒ ﻫﻔﺖ ﺑﻨﺪ( اﺳـﺖ‪ .‬ﺗﯿـﺮه ﭘﻠـﯽ‬ ‫ﺑﯿﻤﺎري ﻫﺎ داﺷـﺘﻪ اﺳـﺖ‪ .‬ﺗﺤﻘﯿﻘـﺎت زﯾـﺎدي روي ﮔﯿﺎﻫـﺎن‬ ‫ﮔﻮﻧﺎﺳﻪ‪ 30،‬ﺟـﻨﺲ و در ﺣـﺪود ‪ 600‬ﮔﻮﻧـﻪ دارد ﮐـﻪ ‪100‬‬ ‫اﻧﺠﺎم ﺷﺪه ﺗﺎ اﺛـﺮات درﻣـﺎﻧﯽ آن ﻫـﺎ ﺑـﻪ اﺛﺒـﺎت ﺑﺮﺳـﺪ‪ ،‬اﻣـﺎ‬ ‫ﮔﻮﻧﻪ ي آن در ﺟﻨﺲ ﮔﯿﺎه روﻣﮑـﺲ )ﺗﺮﺷـﮏ(‪ 180 ،‬ﮔﻮﻧـﻪ‬ ‫‪ / 76‬ﯾﺎﻓﺘﻪ‪ ،‬دوره ﺑﯿﺴﺖ و ﯾﮑﻢ‪ ،‬ﺑﻬﺎر ‪98‬‬ ‫ﺑﺮرﺳﯽ ﻣﮑﺎﻧﯿﺴﻢ اﺛﺮ ﻋﺼﺎره ﻣﺘﺎﻧﻮﻟﯽ ﮔﯿﺎه روﻣﮑﺲ آﻟﻮﺋﻮﻻﺗﻮس ﺑﺮ ﻏﺸﺎء ﮐﺎﻧﺪﯾﺪا ‪...‬‬ ‫ﻏﯿﻮر ﻧﺠﻒ آﺑﺎدي و ﻣﺪﻧﯽ‪‬‬ ‫آن در ﺟﻨﺲ ﭘﻠﯽ ﮔﻮﻧﻮم )ﻋﻠﻒ ﻫﻔﺖ ﺑﻨﺪ( اﺳﺖ‪ .‬اﻧﺘﺸﺎر اﯾـﻦ‬ ‫اﺳــﺘﻔﺎده ﮐــﺮد )‪ .(12‬ﻋﺼــﺎره ﻫــﺎي ﻣﯿــﻮه ﻫــﺎي روﻣﮑــﺲ‬ ‫ﺗﯿﺮه در ﻣﻨﺎﻃﻖ ﺳﺮد ﻧﯿﻤﮑﺮه ﺷﻤﺎﻟﯽ اﺳﺖ‪ 9 .‬ﺟﻨﺲ و ﺑﯿﺶ‬ ‫ﮐﺮﯾﺴﭙﻮس ﻣﺨﺼﻮﺻﺎً در ﺑﯿﻤﺎرﺳﺘﺎن ﻫﺎي ﮐﻮدﮐﺎن ﺑﻪ ﺧـﺎﻃﺮ‬ ‫از ‪ 77‬ﮔﻮﻧﻪ ي اﯾﻦ ﺗﯿﺮه ﺗﻘﺮﯾﺒﺎ در ﺗﻤـﺎﻣﯽ ﻓـﻼت اﯾـﺮان ﺑـﻪ‬ ‫ﻋﻤﻞ ﺑﺎﮐﺘﺮﯾﻮاﺳﺘﺎﺗﯿﮑﯽ و ﺿﺪ اﺳـﻬﺎﻟﯽ ﺧﻔﯿﻔﺸـﺎن ﭘﺬﯾﺮﻓﺘـﻪ‬ ‫ﻃﻮر ﭘﺮاﮐﻨﺪه اﻧﺘﺸﺎر دارﻧﺪ )‪ .(8‬ﺗﺮﺷﮏ ﮔﯿﺎﻫﯽ ﺷﺒﯿﻪ اﺳﻔﻨﺎج‬ ‫ﺷﺪه و ﺑﻪ ﮐﺎر ﺑﺮده ﻣﯽ ﺷﻮد )‪ .(10‬روﻣﮑﺲ ﺳﯿﭙﺮﯾﻮس ﺑﺮاي‬ ‫ﺑﻮده و ﺑﻮﻣﯽ ﻗﺎره اروﭘﺎ اﺳﺖ‪ .‬ﺑﻪ ﻃﻮر وﺣﺸﯽ در ﭼﻤﻦ زارﻫﺎ‪،‬‬ ‫درﻣــﺎن ﺑﯿﻤــﺎري ﻫــﺎي ﭘﻮﺳــﺘﯽ از ﺟﻤﻠــﻪ درﻣﺎﺗﻮﻓﯿﺘﻮزﻫــﺎ و‬ ‫ﮐﺸﺖ زارﻫﺎ‪ ،‬ﮐﻨﺎر رودﺧﺎﻧـﻪ ﻫـﺎ و ﺟـﺎده ﻫـﺎ رﺷـﺪ ﻣﯿﮑﻨـﺪ‪.‬‬ ‫ﻓﻌﺎﻟﯿﺖ آﻧﺘﯽ ﺑﺎﮐﺘﺮﯾـﺎل ﺑـﺮ ﺿـﺪ اﺳـﺘﺎﻓﯿﻠﻮﮐﻮﮐﻮس آرﺋـﻮس‬ ‫ﮐﺮﺑﻮﻫﯿﺪرات ﻫﺎ‪ ،‬ﻓﯿﺒﺮ‪ ،‬ﭼﺮﺑﯽ‪ ،‬ﭘﺮوﺗﺌﯿﻦ‪ ،‬وﯾﺘـﺎﻣﯿﻦ ‪B2 ،B1‬‬ ‫ﻣﻘﺎوم ﺑﻪ ﻣﺘﯽ ﺳﯿﻠﯿﻦ دارد‪ .‬ﻫﻤﭽﻨﯿﻦ ﻣﺸﺨﺺ ﺷـﺪه اﺳـﺖ‬ ‫و ‪
